Kinetic screening is a crucial aspect of the drug discovery process, from confirmation of binding in early discovery to fine-tuning of drug-like properties in pre-clinical development. Grating coupled interferometry (GCI), is a surface-based, label-free biosensing technique. When target molecules (e.g. proteins) are attached to the sensor surface, binding of analytes leads to an increase in surface mass and hence to a change in the refractive index within the evanescent field near the sensing surface. In GCI, refractive index changes on a sensor surface are measured as time-dependent phase-shift signals (interferometry).
In biopharmaceutical applications, GCI plays a pivotal role in characterizing molecular interactions, providing insights on kinetic profiles and binding affinity. The waveRAPID (Repeated Analyte Pulses of Increasing Duration) assay allows for an increase in throughput while reducing overall experimental time.
